How it Works:
Paracetamol has a central analgesic effect mediated through activation of descending serotonergic pathways. Its primary site of action is debated—it may inhibit prostaglandin synthesis or act through an active metabolite influencing cannabinoid receptors.
Usage and Safety
Dosage: Paracetamol (dose as prescribed by physician)
Side Effects:

Neurological: Dizziness, headache, dystonia

Gastrointestinal: Vomiting, dry mouth, diarrhea, constipation, nausea, dyspepsia, enlarged abdomen, gastrointestinal disorder (NOS)

Haematological: Anemia, post-operative hemorrhage

Skin and Appendage: Injection site pain, injection site reaction
Drug Interactions:
Probenecid, barbiturates, isoniazid, anticoagulants, zidovudine, amoxicillin + clavulanic acid, carbamazepine, ethanol, warfarin, phenytoin, busulfan, salicylamide.
Indication:
For relief of mild to moderate pain and reduction of fever when intravenous administration is clinically necessary.
When Not to Use:

History of hypersensitivity to paracetamol or excipients.
Precautions:

May cause serious skin reactions (AGEP, SJS, TEN) which can be fatal.

Discontinue immediately if rash or hypersensitivity signs appear.
Warnings:

Liver Risk: Higher risk in hepatic insufficiency, chronic alcoholism, chronic malnutrition, or dehydration.

Use with Caution: Hepatocellular insufficiency including Gilbert’s syndrome.

Nutritional Concerns: Caution in anorexia, bulimia, cachexia, or chronic malnutrition (low hepatic glutathione).
